Молекулярные механизмы регуляции функциональной активности клеток крови: совершенствование таргетной противовоспалительной терапии

Частота осложнений, связанных с развитием острого воспаления и рецидивированием хронических заболеваний, диктует необходимость совершенствования традиционных методов профилактики и лечения. Перспективным направлением разработки таргетной терапии является управление механизмами активации и рекрутирования лейкоцитов из циркулирующей крови в очаг воспаления, что позволило бы приблизиться к возможности контролированного развития воспалительной реакции. В обзоре приводятся многочисленные доказательства модуляции функциональной активности лейкоцитов и их трансэндотелиальной миграции при стимуляции α- и β-адренорецепторов. Обсуждается практическая значимость экспрессии пуринергических Р2-рецепторов лейкоцитов. Представлены возможные механизмы активации моноцитов и макрофагов при стимуляции Р2Х-рецепторов (P2Х2, P2Х4, Р2Х5 и P2Х7), что позволяет прогнозировать их противовоспалительный потенциал, а в перспективе – контролировать развитие воспаления. Анализируются регуляторные возможности основных подтипов Р2Y-рецепторов (Р2Y1, Р2Y2, Р2Y4, Р2Y6, Р2Y11, Р2Y12, Р2Y13 и Р2Y14), которые участвуют в регуляции фагоцитоза, секреции цитокинов, адгезии и миграции лейкоцитов. Рассматривается роль тромбоцитов, экспрессирующих пуриновые рецепторы (Р2Y1, Р2Y12, Р2Y14 и P2Х1), в рекрутировании и хемотаксисе лейкоцитов при воспалении; обоснована перспективность селективного воздействия на адрен- и пуринергические рецепторы клеток крови как на терапевтическую мишень при системной воспалительной реакции.
